Reference Lab  LabCorp of America
Reference Lab Code  480327 DNA Ploidy, Paraffin Block
Clinical Use  DNA ploidy analysis measures the chromosome content of the cancer cells. Diploid tumors have chromosome numbers of 46 chromosomes. Tumors that are not diploid suggest a major chromosomal disruption that serves as a poor prognostic sign.
Reference Range
Critical Value
Testing Sample Type  PARAFFIN BLK OR 4 (4) micron unstained sections and 2 recut slides or H&E slide
Min Lab Testing Volume  One paraffin block
Special Handling  Specimen Container: Formalin fixed paraffin embedded tissue Formalin fixed paraffin embedded tissue block (preferred) Paraffin Block Bag (preferred) 50 micron unstained sections Formalin fixed, paraffin embedded tissue block Paraffin Block Bag Transport Temperature: Room temperature
Lab Notes  Specimen should be fixed in 10% neutral-buffered formalin. Optimum fixation time is 12 to 24 hours, not to exceed 48 hours.
Methodology  FC - Flow cytometry
Limitations  Use of fixatives other than formalin may not yield equivalent or satisfactory results. No % S phase is reported for tumors analyzed by image cytometry. Small biopsies require image analysis of a 6 u tissue section. This test is not a diagnostic test for cancer.
